Operation of Polarimeter

The purpose of
this SOP is to lay down operational procedure of Polarimeter.

Precautions
7.1
Wear mask and
gloves while operating the instrument.
7.2
Check that no air
bubble is present and the solution is not turbid.
7.3
Before starting the
test wait till sodium lamp warms up.
8.0
Procedure
8.1
Switch on the
instrument using “Power” button.
8.1.1
Wait till sodium
lamp warm and intensity of light reach its maximum for about 15 minutes.
8.1.2
Fill the tube
with distilled water/blank carefully so that no bubble form and place the lens.
8.1.3
If
any bubble is present transfer it to the bulb and place the cell in the chamber
cover it and observe the dark and light band rotate the knob below the eye
piece check for zero.
8.1.4
Take out the
observation tube, discard the blank and rinse it three to four times with the
sample solution.
8.1.5
Now fill the tube
with sample solution carefully to prevent bubble formation and place the lens.
8.1.6
Again
watch through eye piece and match the difference of two bands by rotating the
knob clockwise or anticlockwise.
8.2
Note
the reading where difference got minimum i.e two halves become same.
8.3
Put
the value in the formula and get the results.

8.3.1
If
the knob is rotated clockwise from zero reading will be (+) degree and if it is
anticlockwise from zero then it will be subtracted from 180 and a sign (-) will
be written with value.
8.4
Record
the readings in the Polarimeter log book.
8.5
Switch
off and unplug the instrument and clean properly.
