Lansoprazole Quantitative Analysis On UV Visible Spectrophotometer

¨  By Applying the below method of testing of Lansoprazole we can Analyze active raw material, and different formulated dosage forms.

¨  Absorbance will be measured at  292 nm using UV-Visible Spectrophotometer.

¨  Solvent Used 0.1 M NaOH

¨  Standard Preparation: Accurately weight Lansoprazole working standard 50mg using butter paper and transfer it to a 100ml volumetric flask.

¨  Add 20ml of 0.1M NaOH and sonicate for 10 minutes along with heating at about 80◦C. allow to cool and hen make up the volume with 0.1M NaOH up to 100ml.

¨  Further take 1 ml of this solution into 50ml volumetric flask and make up the volume up to 50ml with 0.1M NaOH .

¨  Sample Preparation: Accurately weight sample equivalent to 50 mg of Lansoprazole and transfer it to a 100ml volumetric flask.

¨  Add 20ml of 0.1M NaOH , first sonicate for 20 minutes along with heating at 80◦C,  and then stir for further 20 minutes. Allow to cool and make up the volume with 0.1M NaOH up to 100ml.

¨  Centrifuge it at 6000 RPM for 5 minutes. Pass the supernatant through filter paper.

¨  Further take 1ml of this solution into 50ml volumetric flask and make up the volume up to 50ml with 0.1M NaOH .

¨  In case of granules and tablets it is necessary to crush the sample and make fine powder sample to attain better solubility.

¨  Measure the absorbance of both sample and standard solutions in a 1 cm quartz cell on a U.V/ Visible spectrophotometer at 292 nm.

¨  Calculation:

¨  %age = Abs. of sample  x conc. of stand x 100

                      Abs. Standard  x conc. of sample
